The Screening Protocol for Selection of Phenodeviants and Results of Large-scale ENU Mutagenesis

Abstract
We tried to develop screening method and searched for novel and various mutants as moldel animals in ENU mutagenesis. After 15mg/kg, 200mg/kg, and 400mg/kg ENU injection to 83 BALB/c Males, which were mated with 605 normal BALB/c FEMALES (6~12 FEMALE MICE PERMALE), 2,066 G_1 and 5,763 G_3 mice were obtained. All produced mice were observed five times from birth to weaning according to ECPS (Esaki and Cho¢¥s Phenotype Screening) protocol which was developed by modification of Irwin¢¥s observation batteries : birthday, 4th, 7th, 14th and 21st day and observation items included development or growth (7 items), morphology (48 items) and behavior (17 items). Total screened mice were 12,279. As the results of screening, we got 157 phenodeviants in G_1 569 phenodeviants in G_3. Fifty nine G_1 phenodeviants were tested for inheritance and 5 dominant mutants were discovered. Twenty two recessive mutants also were found in tested 79 G_1 progenies Early-onset mutant mice included the dysmorphology of face, eye, tail, limb, skin, and foot and abnormal behavior like circling, swimming, head tossing, stiff-walking, and tremor etc. In this study we could effectively screen G_3 recessive mutants. The frequent and concise early-onset screening before weaning will be available for ENU mutagenesis.
